The Effect of Short-term Jumping Load on the Microstructure and Expression of Inflammatory Factors of Bone-patellar Tendon Junctions in Rabbits

    Abstract:

    Objective To investigate the adaptive response of patella patellar tendon junction (PPTJ) and tibial patellar tendon junction (TPTJ) in rabbits under short-term jumping load. Methods Nineteen 18 week old female New Zealand white rabbits were randomly divided into control group (CON, n = 4) and jump 1 day (J1), 3 days (J3), 5 days (J5) group (each group n = 5). Electrical stimulation quantitative jumping device was used to make rabbits complete 150 jumps per day (10 groups × 15 jumps, 3 minutes per group, 6 minutes rest after the 4th and 8th groups, 3 minutes rest after the other groups, the average jumping speed is 0.083 times/ s, the total training time per day is about 70 minutes), the animals were sacrificed 24 hours after the training. The corresponding tissues of the left and right hind limbs were taken for H&E staining and immunohistochemistry. The histomorphological results were qualitatively described, and the cell density, cartilage thickness, and the expression of IL-1β, IL-6, and TGF-β1 were quantitatively monitored. Results The micro-injury of PPTJ and TPTJ occurred in different degrees in the jump groups; the expressions of IL-1β and IL-6 in PPTJ of the J5 group were significantly higher than the CON and J1 groups; the expression of IL-6 in PPTJ of the J3 group was significantly higher than the CON group; the thickness of TPTJ cartilage band in the J5 group was significantly greater than the CON, J1, and J3 groups;in the CON group, the expressions of IL-1β and IL-6 in PPTJ were significantly lower than TPTJ; in the J1 group, the expressions of IL-1β, TGF-β1, IL-6 in PPTJ were significantly lower than TPTJ; in the J3 group, the expression of IL-6 in PPTJ was significantly lower than TPTJ. Conclusions The pathological changes of patella patellar tendon junction induced by the short-term jumping load are earlier than those of tibial patellar tendon junction. The inflammatory factors IL-1β and IL-6 are up-regulated in the process of patella patellar tendon junction injury under short-term jumping load, but there is no significant change of inflammatory factors in tibial patellar tendon junction.
